I had a helluva time finding this, so now that I have I'm blogging it in hopes that it will help others.
C:\Users\<username>\AppData\Roaming\Microsoft\Signatures
UPDATE: Wanted to add something here since this post is probably the most popular one on my blog (possibly next to the "I FUCKING HATE VISTA" one). The location is actually:
%USERPROFILE%\AppData\Roaming\Microsoft\Signatures
Where %USERPROFILE% is an environment variable (you can see what this is by opening a command prompt and typing "set"). You can also just copy/paste the above path into your Explorer address bar and it'll open to that folder. In 99.9% of cases it'll be what's listed above, but if you've got a sneaky sysadmin or you just like to fuck with environment variables then it could be in a location other than C:\Users.

Helped me - Thanks! I did have to turn on "show hidden files and preferences" in "folder options" in the control panel though. Otherwise the AppData folder is hidden.
You don't actually have to show hidden files. You can just type the path into the address bar in Explorer and it'll autocomplete the path, even if the folders are hidden.
